为探究5-羟色胺2A受体(5-HT 2A receptor, 5-HT2AR)基因在海水贝类生长发育中的作用,本研究通过RACE技术和实时荧光定量PCR(qRT-PCR)克隆了厚壳贻贝5-HT2AR 基因的cDNA全长,并分析该基因的时空表达。结果显示,5-HT2AR基因全长2 636 bp,开放阅读框(ORF)2 124 bp,共编码707个氨基酸。序列分析结果显示该序列与人、小鼠、斑马鱼、长牡蛎和虾夷扇贝等物种分别具有45%、45%、48%、49%和67%的同源性。厚壳贻贝雌雄成体各组织和器官中均有5-HT2AR基因表达,雄性中的鳃表达量最高,而在雌性鳃、外套膜和性腺中的表达稍高于其他组织和器官;推测该基因可能与厚壳贻贝的摄食、对外界环境的感知及促进卵母细胞成熟有关。5-HT2AR基因在厚壳贻贝各发育阶段均有表达,且稚贝的表达量为眼点幼虫的1.4倍,推测5-HT2AR基因可能参与了调控厚壳贻贝幼虫的生长发育过程。本研究为进一步了解5-HT基因家族在双壳贝类中的功能奠定了基础。 相似文献

本试验以表达猪乳铁蛋白肽的重组屎肠球菌(pNZ8112-PLFcin/Ef)饲喂断奶仔猪,研究其对仔猪生长性能的影响和抗产肠毒素大肠杆菌(enterotoxigenic Escherichia coli,ETEC)感染的效果。选取28日龄体重相近的健康断奶仔猪36头,随机分为3组(重组屎肠球菌组、空载体组和培养基组),每组3个重复,每个重复4头仔猪。重组屎肠球菌组和空载体组分别饲喂添加pNZ8112-PLFcin/Ef (6×1012 CFU/kg)和pNZ8112/Ef (6×1012 CFU/kg)的基础日粮,而培养基组饲喂含有相同体积的GM17液体培养基的基础日粮。试验期26 d。结果显示,与培养基组相比,重组屎肠球菌组断奶仔猪的平均日增重极显著提高(P<0.01);料重比显著降低(P<0.05);腹泻率明显降低,肠道菌群的均匀度和多样性指数均下降。为进一步探究表达猪乳铁蛋白肽的重组屎肠球菌对断奶仔猪抵抗ETEC感染的保护作用,在连续饲喂21 d后,每个重复中随机挑选1头体况相近的断奶仔猪灌服ETEC。结果发现,与培养基组相比,攻菌后重组屎肠球菌组断奶仔猪血清白细胞介素-2(IL-2)、免疫球蛋白G (IgG)含量、肠黏液中分泌型免疫球蛋白A (sIgA)水平均显著升高(P<0.05);脾脏指数显著提高(P<0.05),但胸腺指数、肠段长度及重量则均无显著差异(P>0.05)。综上所述,表达猪乳铁蛋白肽的重组屎肠球菌能够起到促进断奶仔猪生长及抗ETEC感染的保护作用。 相似文献

Marine fisheries target and catch fish both for direct human consumption (DHC) as well as for fishmeal and fish oil, and other products. We derived the fractions used for each for 1950–2010 by fishing country, and thus provide a factual foundation for discussions of the optimal use of fisheries resources. From 1950 to 2010, 27% (~20 million tonnes annually) of globally reconstructed marine fisheries landings were destined for uses other than DHC. Importantly, 90% of fish destined for uses other than DHC are food‐grade or prime food‐grade fish, while fish without a ready market for DHC make up a much smaller proportion. These findings have implications for how we are using fish to feed ourselves or, more appropriately, how we are not using fish to feed ourselves. 相似文献

We evaluated the effects of Bacillus cereus, as an additive in water and feed, on the gut microbiota and immunological parameters of Nile tilapia (Oreochromis niloticus) fingerlings. Experiments were performed in tanks and net cages respectively. Experiment 1: Tilapia were housed in tanks for 42 days, and B. cereus was added to the water at 1.0 × 104 cfu mL?1 (Treatment 1) and 1.0 ×105 cfu mL?1 (Treatment 2) weekly. For the control, no probiotic was added. Experiment 2: Tilapia were housed in cages for 42 days, and the feed was supplemented with B. cereus at 1.0 × 107 cfu g?1 (Treatment 1) and 1.0 × 108 cfu g?1 (Treatment 2) weekly. For the control, no probiotic was added. Each treatment contained three replicates, with 50 male tilapias per replicate. The fish from the probiotic treatments in both tank and cage experiments had significantly higher serum lysozyme and peroxidase activities than the control. In the cage experiment, alkaline phosphatase and total superoxide dismutase activities in tilapia were significantly higher in probiotic treatments compared with the control. The results of polymerase chain reaction‐denaturing gradient gel electrophoresis showed that B. cereus supplementation in the feed and water affected the autochthonous gut bacteria community of tilapia and stimulated various potentially beneficial bacteria. Therefore, B. cereus, as a water or feed additive, could enhance the immune status and affect the gut microbiota of tilapia. 为了调节循环水系统中养殖水体的pH,根据气体交换原理,设计一种脱二氧化碳(CO_2)装置。采用该装置去除养殖水体中的CO_2,并对由于CO_2含量累积造成的pH下降进行调节,使养殖鱼类处在一个适宜的pH环境中。试验时水温控制在(25±0.5)℃,每1 h取水样测1次pH,每4 h测1次碱度。水样取自养鱼桶内的水,检测前先对水样用40μm孔径针头过滤器进行过滤处理,实验周期24 h。结果显示,循环水系统加装脱二氧化碳装置能有效去除CO_2,使水体稳定在一个适宜的pH范围(7.39~7.42);CO_2质量浓度呈降低趋势,24 h后由开始的13.16 mg/L降低到7~8 mg/L,降低近50%,而不加装脱二氧化碳装置的循环水系统CO_2质量浓度持续上升,24 h后增加到37 mg/L左右,pH持续降低,最终降低到6.72~6.81。研究表明,脱二氧化碳装置能够有效去除水体中的CO_2,使水体pH维持在一个适宜鱼类生长的范围。 相似文献

Although stable isotope analysis is a powerful tool for determining diet, migration patterns and the structure of food webs in aquatic systems, the slow response of isotopic ratios in the widely used muscle tissue often hampers this approach, particularly in slow‐growing or adult fishes. We conducted a diet‐switch experiment to compare the changes in the stable carbon isotope ratio (δ13C values) in the epidermal mucus and muscle tissue of five‐year‐old catfish (Silurus asotus). The isotope ratios in noninvasively sampled mucus changed more rapidly than those in the muscle tissue. As isotopic change in mucus was relatively rapid, this technique can be used over a finer timescale than traditional isotopic analyses using the muscle tissue. The isotopic change half‐life in our experimental condition was 200 days, which would not be short enough for some research purposes. Examining mucus along with the muscle would enable food habits of slow‐growing fishes to be determined over different timescales. However, the rate of isotopic change in the mucus was negatively affected by the size of fish and was slower than the previously reported rates in juvenile steelhead. These findings suggest that mucus turnover rates need to be determined prior to the field data interpretation. 相似文献

This study aimed at elucidating SS-bonds of HMW-gliadins (HGL) from wheat with the focus on terminators of glutenin polymerisation. HGL from wheat flour extracts non-treated or treated with the S-alkylation reagent N-ethylmaleinimide (NEMI) were compared. HGL from wheat flour Akteur were isolated, hydrolysed with thermolysin and the resulting peptides pre-separated by gel permeation chromatography and analysed by liquid chromatography/mass-spectrometry using alternating electron transfer dissociation/collision-induced dissociation. Altogether, 22 and 28 SS-peptides from samples without and with NEMI treatment, respectively, were identified. Twenty-six peptides included standard SS-bonds of α- and γ-gliadins, high-molecular-weight and low-molecular-weight glutenin subunits. Eleven SS-bonds were identified for the first time. Fifteen peptides unique to HGL contained cysteine residues from gliadins with an odd number of cysteines (ω5-, α- and γ-gliadins). Thus, gliadins with an odd number of cysteines, glutathione and cysteine had acted as terminators of glutenin polymerisation. Decisive differences between samples without and with NEMI treatment were not obvious showing that the termination of polymerisation was already completed in the flour. The two HGL samples, however, were different in the majority of ten peptides that included disulphide-linked low-molecular-weight (LMW) thiols such as glutathione and cysteine with the former being enriched in the non-treated HGL-sample. 相似文献

【目的】香稻作为一类特殊的水稻群体,以其清香可口的品质特性备受消费者的欢迎。到目前为止,水稻中的香味主要受第8染色体上编码甜菜碱醛脱氢酶基因Badh2控制。【方法】通过CRISPR/CAS9技术对中花11的香味基因Badh2进行编辑。【结果】获得转基因T_0代植株并对其所衍生的T_1代20个单株进行了鉴定分析,获得了一个剔除了载体骨架且第1外显子上插入一个碱基T的突变体材料。该材料中Badh2 RNA水平显著下调;利用GC-MS技术测定野生型及突变体材料籽粒2-乙酰-1-吡咯啉含量,结果表明突变体材料中的香味物质显著增加;此外,我们还对野生型及T_2代香型植株水稻产量及稻米蒸煮食味品质进行了考查及测定分析,发现除分蘖数及结实率呈现出显著差异外(P0.05),其余各项指标在两组材料间都无显著差异。【结论】通过CRISPR/CAS9技术成功地对水稻香味基因进行了编辑,可为香稻育种提供丰富的理论指导,加快香稻的育种进程。 相似文献
